What the research says about LGD-4033
The single most useful paper on this compound is the phase I study by Basaria and colleagues, published in the Journals of Gerontology Series A in 2013. Seventy six healthy young men received placebo or LGD-4033 daily for three weeks, at doses used in that published research of 0.1mg, 0.3mg and 1mg. Lean body mass rose in a dose dependent fashion, reaching roughly 1.2kg above placebo in the 1mg group over just twenty one days, with no change in prostate specific antigen. The compound was well tolerated over that short window.
The same study recorded the findings that responsible discussion should always carry alongside the lean mass figure. Total testosterone, sex hormone binding globulin, free testosterone, HDL cholesterol and triglycerides all fell in a dose dependent manner, and follicle stimulating hormone was suppressed. Hormone levels returned towards baseline during follow up after the three week course ended. A later phase II programme in patients recovering from hip fracture reported increases in lean body mass, but the full data set has never been published in a peer reviewed journal.
Mechanistically LGD-4033 is a high affinity, high selectivity ligand for the androgen receptor with an oral bioavailability and half life, around 24 to 36 hours in the published pharmacokinetic work, that suit once daily administration in a study protocol. It binds the receptor considerably more strongly than ostarine does, which explains why the suppression of endogenous hormones appears at much lower milligram figures. Published case reports have described liver injury in people who took products sold as LGD-4033, and in the cases where the product was analysed the contents frequently did not match the label.
