RAD-140, usually called testolone, is a non steroidal selective androgen receptor modulator discovered at Radius Health, which later investigated it in androgen receptor positive breast cancer. RAD-140 was introduced to the literature by Miller and colleagues in ACS Medicinal Chemistry Letters in 2011. That paper describes the discovery chemistry and the preclinical work: in castrated rats the compound produced anabolic activity in levator ani muscle comparable to testosterone while leaving the prostate largely unaffected, and in a primate study it increased lean body mass over twenty eight days. The selectivity ratio reported in that work is the origin of almost everything written about the compound since.
Subsequent academic interest has run in two directions. Jayaraman and colleagues reported in Endocrinology in 2014 that RAD-140 had neuroprotective activity in cultured rat neurons and in a rat model, work that treated the compound as a possible androgen replacement for the brain rather than for muscle. Separately, Radius Health investigated RAD-140 in androgen receptor positive breast cancer, and a phase I trial in that indication was registered. No peer reviewed trial has reported lean mass or strength outcomes for RAD-140 in healthy human volunteers, which is the central gap in its evidence base.
The safety literature that does exist in humans comes from case reports rather than trials. Several papers in hepatology and internal medicine journals since 2019 have described cholestatic or mixed pattern liver injury in people who took products sold as RAD-140, sometimes alongside other compounds and often without any analysis of what the product actually contained. Preclinical work also shows the expected suppression of endogenous androgen production. Anyone reading about this compound should weigh a strong preclinical record against a thin and largely adverse human record.

Is RAD-140 legal in the UK in 2026? SARMs are not controlled substances under the Misuse of Drugs Act 1971, so buying and possessing RAD-140 as a research chemical is lawful in the United Kingdom in 2026. What is not lawful is selling or advertising it for human use, because the MHRA treats it as an unlicensed medicine in that context. Is RAD-140 prohibited in sport? Yes, at all times, under section S1.2 of the WADA prohibited list. Testolone has featured in a number of published anti doping decisions and is detectable by routine screening. It should be assumed to be reportable at trace concentrations.
What makes RAD-140 different from RAD-150? RAD-150, also sold as TLB-150, is the benzoate ester of RAD-140. Esterification is a formulation strategy intended to change stability and release rather than to change the underlying receptor pharmacology. The published evidence sits almost entirely with RAD-140 itself; RAD-150 has no meaningful literature of its own.
