AOD-9604, also written AOD9604 or AOD 9604 and often searched simply as AOD peptide, is a 16 amino acid peptide made from the C-terminal end of human growth hormone (residues 177 to 191 with an added tyrosine), developed to study whether the fat metabolism activity of growth hormone could be separated from its effects on growth. What the research says about AOD-9604
The compound came out of work at Monash University in Melbourne in the 1990s, which found that a small fragment of growth hormone appeared to carry much of the hormone's effect on fat tissue without its effect on blood glucose or IGF-1. Metabolic Pharmaceuticals developed the stabilised form as AOD-9604, and Heffernan and colleagues reported in Endocrinology in 2001 that it reduced body weight gain and fat in obese mice without the growth promoting activity of the full hormone.
The human programme is where the story changes. Early phase trials suggested the peptide was well tolerated, but the larger phase IIb trial completed in 2007, in roughly 500 people over 24 weeks, did not show a meaningful difference in weight loss compared with placebo, and development for obesity was stopped. That result is the most important fact about AOD-9604 and is routinely left out of retail descriptions.
Research interest has since moved elsewhere. Kwon and Park reported in Annals of Clinical and Laboratory Science in 2015 on AOD-9604 in a rabbit model of osteoarthritis, examining cartilage alongside hyaluronic acid. AOD-9604 remains a useful reference compound for laboratory work on growth hormone fragments and lipid metabolism, with a well documented safety record in early trials and a negative efficacy result in its main one.
