NAD+ (nicotinamide adenine dinucleotide) is a coenzyme present in every living cell, central to redox reactions, energy metabolism and the activity of sirtuins and PARP enzymes. Cellular NAD+ levels decline with age in animal and human tissue studies.

The literature is large and mostly concerns NAD+ precursors (nicotinamide riboside and nicotinamide mononucleotide) taken orally: Martens and colleagues reported that nicotinamide riboside raised blood NAD+ in older adults (Nature Communications, 2018), and Yoshino and colleagues reported effects of NMN on muscle insulin sensitivity in prediabetic women (Science, 2021). Direct NAD+ has been studied by intravenous infusion in small clinical settings, for example Grant and colleagues (Frontiers in Aging Neuroscience, 2019) who measured plasma NAD+ and metabolites during infusion.
